Pain resolution in bortezomib-induced polyneuropathy: the role of the blood nerve barrier and claudin-11 - FR93

Bortezomib-induced peripheral neuropathy (BIPN) is challenge in the treatment of multiple myeloma because factors governing prevention and recovery as well as pain resolution are unclear. In our study, we analyzed the nerve physiology in a preclinical model of BIPN in rats at the time points corresponding to maximum hypersensitivity and pain resolution. We discovered a novel role of the tight junction protein Cldn11 as it was localized in the perineurium and Schwann cells, which explained perineurial leakiness and axonal swelling. Ntn1 was confirmed as a possible master regulator of perineurial barrier stability and axonal regeneration in the skin.
